Methylfolate in Oral Pouches
Methylfolate is the folate form some energy and focus pouches use, and one of the easiest vitamins to get wrong on the label. This guide covers salt forms, dose math against the upper limit, the unsettled DFE conversion, stability, and testing.
What methylfolate is
Methylfolate, or L-5-methyltetrahydrofolate (L-5-MTHF), is a form of folate, which is a vitamin under 21 U.S.C. 321(ff)(1)(A). The NIH Office of Dietary Supplements (ODS) explicitly recognizes it as a supplement form of folate. It is sold as a salt, and the salt matters: the calcium salt has a USP monograph ("Calcium L-5-Methyltetrahydrofolate"), while the glucosamine salt does not. The adult Daily Value is 400mcg DFE (dietary folate equivalents).
A common misconception is that methylfolate has no upper limit because it is the "natural" form. ODS set the 1,000mcg per day adult UL for the synthetic forms of folate in supplements and fortified foods, and supplemental L-5-MTHF is one of them. Only folate from food has no UL.
Why brands put methylfolate in a pouch
It rounds out a B-vitamin panel at a trivial cost to the formula. PouchMade's standard pouch is 0.6g, with 0.5g and 0.7g formats available, and a typical actives budget of 250-280mg per pouch, about 300mg at most; filler, flavor, and sweetener make up the rest. 400mcg is 0.4mg, about 0.14 to 0.16 percent of a 250-280mg budget. Capacity is not the issue; one pouch can carry a full Daily Value, and methylfolate is a supporting inclusion.
The issue is the daily total. At 400mcg of methylfolate per pouch, two and a half pouches reach the 1,000mcg UL. At 100mcg per pouch it takes ten. One fully disclosed formula in the market handles this well: 100mcg DFE per pouch, so its stated four-pouch daily maximum lands exactly at 400mcg DFE, 100 percent of the Daily Value. That is the right design logic: size the pouch to the directed daily maximum, not to a single serving.
Vitamins are largely carried by saliva and swallowed, and there is little or no pouch-specific absorption data for methylfolate. The pouch is a convenience format.
Industry-typical dose ranges
In a PouchMade review of about 36 non-nicotine pouch formulas on major US marketplaces in September 2026, 2 used folate, and both disclosed an exact dose: 100mcg DFE per pouch as L-methylfolate, and 675mcg DFE per pouch (170 percent of the Daily Value) as folic acid. Only one formula used methylfolate specifically. A wider scan of about 198 pouch products found two more brands stating folate only as a percent Daily Value (170 percent and 100 percent) with no microgram figure. Folate shows up in caffeine plus B-vitamin support blends, beside B12, B6, and niacin, and in focus formulas.
| Reference point | Amount |
|---|---|
| Adult Daily Value | 400mcg DFE |
| Adult UL, supplemental folate, per day | 1,000mcg |
| Disclosed pouch doses, September 2026 | 100-675mcg DFE per pouch |
| Share of a 250mg actives budget at 400mcg | 0.16% |
Formulation: taste and mouthfeel
At microgram levels methylfolate has no practical effect on taste. The formulation work is stability and uniformity. L-5-MTHF is sensitive to oxidation, heat, and light. Published work (Molecules, 2021) found the crystalline calcium salt more stable than the amorphous form or the glucosamine salt, and a post-market comparison agreed; ascorbic acid and cysteine protect it in solution, a point in favor of pairing it with vitamin C. Supplier stability claims conflict, so run your own accelerated stability on the finished pouch. For heat-sensitive folates, 30 C / 65 percent RH data can be more honest than 40 C data, which may fail for reasons that would never occur at room temperature.
Uniformity is the other half. 100mcg in a 600mg fill is under 0.02 percent of the pouch, so it needs a controlled blending step and a finished-product test that proves it. Added vitamins are Class I nutrients under 21 CFR 101.9(g) and must stay at 100 percent or more of label through shelf life; any overage should be justified by stability data and counted against the UL.
Quality and testing
Identity and assay run by HPLC against a reference standard, and the standard has to match the salt actually purchased. For the calcium salt, the USP monograph sets:
- Assay: not less than 95.0 percent and not more than 102.0 percent.
- Isomer purity: d-isomer not more than 1.0 percent, which a CoA should report as a number.
- Heavy metals: lead, arsenic, cadmium, and mercury by ICP-MS, to USP <2232> limits.
- Microbial: counts per USP <2021>, <2022>, and <2023>.
Per 21 CFR 111.75, identity is tested on every incoming lot; a supplier CoA does not replace it.
Regulatory notes
Folate is a vitamin, but New Dietary Ingredient status is decided form by form, and there is no FDA list of "old" dietary ingredients. Document pre-1994 marketing or an existing NDI notification for the specific 5-MTHF salt in the formula; the basis for the glucosamine salt should be confirmed before relying on it.
Labeling is unsettled. ODS states that factors for converting micrograms of 5-MTHF to micrograms DFE have not been formally established, so confirm FDA's current position with regulatory counsel before setting the Supplement Facts panel. A wrong DFE conversion is a common error. Claims stay structure/function with the DSHEA disclaimer, and directions keep "swallow saliva."
Where methylfolate fits at PouchMade
Methylfolate belongs in a B-vitamin panel in an energy pouch or a functional pouch. PouchMade formulates custom stacks through custom formulation at its FDA-registered, GMP-certified facility in Dallas, Texas, producing dietary supplements under 21 CFR 111. The first questions are which salt, how many pouches a day the label directs, and how the panel will declare DFE.
Q01Does methylfolate have an upper limit?
Yes. The 1,000mcg per day adult UL applies to synthetic folate forms in supplements and fortified foods, and supplemental L-5-MTHF is one of them. Only folate from food has no UL.
Q02How much methylfolate is in a pouch?
In a September 2026 PouchMade review of about 36 pouch formulas, the one methylfolate formula carried 100mcg DFE per pouch, sized so its four-pouch daily maximum equals the 400mcg DFE Daily Value.
Q03Which form of methylfolate is best for a pouch?
The crystalline calcium salt has a USP monograph and was more stable than the amorphous form and the glucosamine salt in published comparisons. Supplier data conflicts, so confirm with your own stability testing.
Q04How is methylfolate declared on a Supplement Facts panel?
Folate is declared in mcg DFE, but ODS says conversion factors for 5-MTHF to DFE have not been formally established. Confirm the current FDA position with regulatory counsel before finalizing the panel.
